The terminal deoxynucleoside transferase TdT01 is a mesophilic enzyme of eukaryotic origin. This enzyme is a template-independent DNA polymerase which catalyzes the random addition of deoxynucleoside 5'-triphosphate to the 3'-end of a DNA initiator one nucleotide at a time. It cannot initiate a chain de novo. The nucleoside may be ribo- or 2'-deoxyribo-.
