Nucleoside phosphorylases

Nucleoside phosphorylases (NPs) are transferases catalyzing the reversible cleavage of the C–N glycosidic bond by a phosphate group to yield the free nucleobase and ribose-1-phosphate. Based on their substrate spectrum they can be classified into purine nucleoside phosphorylases (PNPs, EC 2.4.2.1) and pyrimidine nucleoside phosphorylases (PyNPs, EC 2.4.2.2).

 

PyNPs catalyze the reversible phosphorolytic cleavage of pyrimidine nucleosides. Natural substrates are both uridine and thymidine. PyNPs of bacterial origin use a wide variety of modified nucleosides as substrates. Cytidine, however, is not converted by pyrimidine nucleoside phosphorylases.

 

PNPs catalyze the reversible phosphorolytic cleavage of purine nucleosides. Natural substrates are both adenine and guanine. PNPs of bacterial origin use a wide variety of modified nucleosides as substrates.

 

By combining a pyrimidine and a purine nucleoside phosphorylase, the enzymatic synthesis of natural and modified nucleosides is possible in a one-pot, two enzyme transglycosylation reaction.
